Both tubes have max Plate Voltage for amps as 300V. The difference of 100V and 250V is only for Class A1 amp, that is where the grid is negative. Not for others. People who do not drink, do not smoke, do not eat red meat will one day feel really stupid lying there and dying from nothing.
